LC-Mass Spectrometry (LC-MS) is a powerful analytical tool for separation, detection and quantitation of analytes in complex mixtures. Liquid chromatography (LC) and mass spectrometry (MS) are selective techniques that when combined have unique analytical capabilities. The LC separates analytes based on their physico-chemical properties while the MS provides high sensitivity and capability to measure low abundance metabolites or markers that may not separate by GC or HPLC. Liquid chromatography-mass spectrometry (LC-MS) is a widely used method for separating, detecting and quantifying compounds.  This lab provides an example of how LC-MS can be used to positively identify molecules, by comparing their mass spectrum with a reference chart.

Luteinizing hormone (LH) is a hormone produced in the anterior pituitary gland involved in regulation of the reproductive function. Specifically, gonadotropic cells in the anterior pituitary gland produce LH or Luteinizing hormone LH SLH production is regulated by gonadotropin-releasing hormone (GnRH) from the hypothalamus. An acute surge in LH levels triggers ovulation and the development of the corpus luteum in females. In males, LH is also called interstitial cell-stimulating hormone (ICSH). They act on the Leydig cells to produce testosterone and work in synergy with the follicle-stimulating hormone (FSH).

The structure of LH hormone is similar to that of FSH hormone. Both LH and FSH are glycoproteins, containing the common heterodimeric subunits. The beta-subunit is similar in both hormones, but the alpha-subunit is different with LH having 92 amino acids and FSH having 97 amino acids.
